New research indicates that individuals suffering from Long COVID may have a significant reduction in dopamine-producing brain cells. This neurotransmitter, dopamine, is vital for regulating movement, memory, motivation, and mood.

Long COVID, or post-COVID-19 condition, describes symptoms that persist or arise at least three months after initial COVID-19 infection, without other explanations. Common complaints include fatigue, cognitive difficulties, memory loss, lack of motivation, and slowed movement.

Study Overview

Led by Jeffrey Meyer and published in eBioMedicine, the study involved 24 adults experiencing Long COVID symptoms and compared them with healthy controls. Participants underwent PET scans using a tracer to measure vesicular monoamine transporter 2 (VMAT2), a protein found predominantly in dopamine-releasing nerve terminals.

The scans revealed approximately an 18% reduction in dopamine nerve terminals among Long COVID patients relative to healthy individuals. Lower dopamine activity correlated with symptom severity, including:

  • Reduced motivation linked to decreased dopamine in the ventral striatum
  • Slower motor function associated with diminished dopamine in the dorsal putamen
  • Memory impairments connected to dopamine deficits in the dorsal caudate

Implications and Limitations

The findings offer insight into the neurological underpinnings of persistent Long COVID symptoms, although the study does not establish direct neuronal destruction by the virus. The research was observational and limited by sample size and focus on neurological symptoms, thus not generalizable to all Long COVID cases.

Importantly, standard blood tests failed to detect these brain changes, highlighting the need for advanced neuroimaging in understanding Long COVID.

Future Directions

The study suggests potential therapeutic strategies targeting dopamine pathways, including drugs that enhance dopamine release or reduce brain inflammation. Clinical trials are necessary to evaluate whether such treatments can improve recovery and quality of life for the millions affected globally by Long COVID.
